Create/Load a channel

create_load_channel

The expert users (if this option is activated) have access to all optical elements of the microscope, wheel filter, dichroic cube and light source. To manually create a fluorescent channel, you should follow the steps below:

  1. Select the right excitation source.
  2. Set its intensity.
  3. Adjust dichroic cubes.
  4. Adjust excitation/emission fiters.
  5. If necessary, select brightfield light instead of or in addition to fluorescence.
  6. Name your channel.
  7. Save it.
The created channel will be available in the list.
Important: Some systems have more than one camera port. It is necessary to select the right one before imaging.
Tip: In User mode, you can modify the channel (excitation, filter, power) and save it for your experiment by clicking on Add. You will be able to recall the modified channel in the Acquisition tab. If you close the interface and reload it, the modified channel will not appear again. In Expert mode, you can modify the channels and save them for future acquisitions.
